miR-381在子宫颈鳞癌患者血清和组织中的表达及意义

Expression and significance of miR-381 in serum and tissues of patients with cervical squamous cell carcinoma

  • 摘要: 目的 探讨miR-381在子宫颈鳞癌患者血清和组织中的表达及意义。方法 选取2013年6月-2018年6月深圳市南山医院收治的50例宫颈鳞癌患者和50例宫颈上皮内瘤变Ⅲ级患者为研究对象,另选取同期在深圳市南山医院因子宫肌瘤行子宫全切术患者50例纳入对照组,采用RT-PCR法分别检测3组血清样本和癌组织样本中miR-381表达水平,并分析miR-381表达水平与宫颈鳞癌患者临床病理特征的相关性。结果 miR-381在宫颈鳞癌组和宫颈上皮内瘤变Ⅲ级组患者血清和癌组织中的相对表达量明显低于对照组,且差异均有统计学意义(P<0.05);miR-381在宫颈鳞癌组患者血清和癌组织中的相对表达量明显低于宫颈上皮内瘤变Ⅲ级组,且差异均有统计学意义(P<0.05);宫颈鳞癌患者血清和癌组织中miR-381的表达与病理分级、宫颈间质浸润、盆腔淋巴结转移均存在相关性(P<0.05),而与患者年龄及FIGO分期无明显相关性(P>0.05);血清中miR-381对宫颈鳞癌的诊断效能ROC曲线下面积为0.821(95%CI:0.597~1.042),诊断敏感性为92.07%,特异性为79.42%;癌组织中miR-381对宫颈鳞癌的诊断效能ROC曲线下面积为0.893(95%CI:0.704~1.625),诊断敏感性为95.82%,特异性为83.46%。结论 miR-381在宫颈鳞癌患者血清和癌组织中均呈低表达,其表达水平与宫颈鳞癌的发生发展存在相关性,可能成为宫颈鳞癌特异性诊断的潜在标记物。

     

    Abstract: Objective To investigate the expression and significance of miR-381 in serum and tissues of patients with cervical squamous cell carcinoma.Methods From June 2013 to June 2018, 50 patients with cervical squamous cell carcinoma and 50 patients with cervical intraepithelial neoplasia grade 3 (CIN3) admitted to our hospital were selected as subjects. Another 50 patients who underwent hysterectomy in our hospital for uterine fibroids were included in the control group. The expression levels of miR-381 in serum samples and tissue samples were detected by RT-PCR, and the correlation between miR-381 expression and clinicopathological features of cervical squamous cell carcinoma was analyzed.Results The relative expression of miR-381 in serum and tissues of patients with cervical squamous cell carcinoma and CIN3 was significantly lower than that of the control group, and the difference was statistically significant (P<0.05). The relative expression of miR-381 in serum and tissues of patients with cervical squamous cell carcinoma was significantly lower than that of CIN3, and the difference was statistically significant (P<0.05). The expression of miR-381 in serum and tissues of patients with cervical squamous cell carcinoma was correlated with pathological grade, cervical interstitial infiltration and pelvic lymph node metastasis (P<0.05), and there was no significant correlation with patient age and FIGO stage (P>0.05). The area under the ROC curve of the diagnostic potency of miR-381 in serum of cervical squamous cell carcinoma was 0.821 (95%CI: 0.597-1.042). The diagnostic sensitivity was 92.07%, and the specificity was 79. 42%. The area under the ROC curve of the diagnostic efficiency of miR-381 in tissues of cervical squamous cell carcinoma was 0.893 (95%CI: 0.704-1.625). The diagnostic sensitivity was 95.82%, and the specificity was 83.46%.Conclusions miR-381 shows low expression in serum and tissues of patients with cervical squamous cell carcinoma, and its expression level is correlated with the occurrence and development of cervical squamous cell carcinoma. miR-381 may be a potential marker for the specific diagnosis of cervical squamous cell carcinoma.
