Abstract:
Objective To explore the influence of Bacille Calmette-Guerin (BCG) vaccine intervention on the expression of macrophage inflammatory cytokines and adhesion molecules in IL-4 induction.
Methods Macrophages were stimulated with PMA (10 ng/ml) and IL-4 (10 ng/ml), and transformed into M2. Then they were dissolved at DMSO 0 mg/L, 20 mg/L, 50 mg/L and 100 mg/L BCG for 24 hours. The expressions of IL-12, IL-6, IL-10, MRC-1 and CCL-2 mRNA in each group were determined by PCR methods.
Results After IL-4 stimulation, the contents of IL-12, IL-6, IL-10, MRC-1 and CCL-2 mRNA were increased significantly (
P<0.01). After dissolved in BCG, the contents of IL-12, IL-6, IL-10, MRC-1 and CCL-2 mRNA were decreased significantly (
P<0.01). It was more significant after dissolved at 100 mg/L BCG.
Conclusion The BCG vaccine can inhibit the expression of the macrophage inflammatory cytokines (IL-12, IL-6, IL-10, MRC-1) and Adhesion molecule (CCL-2) which were stimulated by IL-4.
