Effects of stereoisomers of inositol on insulin sensitivity of gestational diabetic mice and its mechanism

  • Objective To investigate the effects of inositol stereoisomers on insulin sensitivity in gestationally diabetic mice and to explore its mechanism.Methods The model of gestationally diabetes was constructing. The 24-hour changes of water consumption,food intake and body weight of the model were recorded and the random blood glucose and fasting blood glucose were detected to verify the success of the model. The gestationally diabetic mice were randomly allocated into intervention group 1(MI),intervention group 2(DCI),and control group. The weight,water consumption,and food intake of the test animals were dynamically observed. The oral glucose of the animals in the above groups were tested,and the insulin resistance index was calculated and the difference was observed.The changes of hormone secretion were detected by ELISA.Results An animal model of gestationally diabetes mellitus was successfully constructed by injecting streptozotocin(STZ) on the basis of a high-fat diet. Compared with the control group,there were statistically significant differences in 24-hour water consumption,food intake,body weight,random blood glucose,and fasting blood glucose between the model group and the modeling control group(P<0.05). After the intervention of inositol stereoisomers,compared with the control group,the random blood glucose and fasting blood glucose of the intervention group 1 and the intervention group 2 decreased,and the difference was statistically significant(P<0.05). The decrease of intervention group 2 was more obvious.Compared with the control group,the peripheral blood INS,LEP,CRP and ADP of the intervention group 1 and the intervention group 2 were significantly different and had statistically significant(P<0.05). Draw a curve based on the blood glucose values measured at different time. And the K value of intervention group 1(K: intervention group 1 = 49.85 ±4.39) and intervention group 2(K:intervention group 2 = 52.17 ±4.39) compared with control group(K: Control group = 32.87 ±4.39) had significant statistical difference(P<0.05).Conclusion D-chiro-inositol(DCI) can significantly improve insulin resistance and increase insulin sensitivity in gestational diabetic mice.
